The simplest form of that macro is
#showtooltip
/cast [noharm][target=targettarget] Righteous Defense
which will work on NPCs (buffable or not).
As long as [noharm] (or [help], though that won't work directly on NPCs you can't buff) is first, the [target=targettarget] alone works fine.
Specifically, NPCs that are not buffable are not considered "help". NPCs who can be buffed (ala the Prisoner from the Skettis daily) are.
So casting it without the macro will work on the Prisoner, for example.
NPCs are considered "harm" as long as they are hostile, though - and [noharm] specifically works on all non-hostile mobs.
