How do I use the runes? Tried to use one and ended up killing myself.

You set them as traps. You right click on them (in your inventory) to activate, and then left click someplace nearby to place it. Then you do not go near it, but you might lure monsters over it.

I dropped a couple (fire) when I was dealing with the butcher, nothing happened. He just walked right over them.
For now I will continue to play the game and learn how to use them correctly.

Did you drop them or did you right click and get the "casting cursor" before placing them?

I used the right click to set them but nothing happened. So I figure that I must have not done it correctly. Cuz the butcher kept coming at me swinging his beloved cleaver.

Right click to activate, left click where you want them placed to place them - it's just like a town portal scroll.
To be honest, the worst place to do that is around the butcher, what with his speed.
